3-Methyladenine is an inhibitor of class III phosphatidylinositol 3-kinase (PI 3-kinase).It can protect cerebellar granule cells from apoptosis following serum/potassium deprivation. 3-Methyladenine inhibits phosphoinositide 3-kinase (PI3K) activity, which plays a key role in autophagy process.

3-Methyladenine, aslo known as NSC 66389,is a selective PI3K inhibitor for Vps34 and PI3Kγ, blocks class I PI3K consistently, whereas suppression of class III PI3K is transient, and also suppress the formation autophagosome. 3-methyladenine enhances the sensitivity of nasopharyngeal carcinoma cells to chemotherapy and radiotherapy. 3-Methyladenine can depress drug efflux transporters via blocking the PI3K-AKT-mTOR pathway thus sensitizing MDR cancer to chemotherapy. 3-methyladenine potentiates apoptosis induced by dietary tocotrienols in breast cancer cells. 3-MA inhibits autophagy by blocking autophagosome formation via the inhibition of type III Phosphatidylinositol 3-kinases (PI-3K). For use as an autophagy inhibitor, 3-MA is typically used at a concentration of 5 mM. In studies investigating N-methylations of DNA bases, use of deuterated methylating agents allows for monitoring of alkylation patterns with mass spectrometry. 3-Methyl-d3-adenine is potentially useful as a reference sample for identifying adenine alkylations in such studies.
